{"id":60,"date":"2013-07-17T17:36:48","date_gmt":"2013-07-17T21:36:48","guid":{"rendered":"https:\/\/sites.bu.edu\/biomol\/?page_id=60"},"modified":"2013-07-18T17:03:58","modified_gmt":"2013-07-18T21:03:58","slug":"gene-imaging","status":"publish","type":"page","link":"https:\/\/sites.bu.edu\/biomol\/projects\/gene-imaging\/","title":{"rendered":"Gene Imaging"},"content":{"rendered":"<p>We applied our PNA-RCA-FISH approach for \u00a0<i>in situ<\/i> detection of short, single-copy sequences within genomic DNA in human cells.\u00a0We employed this technique for fluorescent imaging of unique target sites on human mitochondrial and autosomal DNA.\u00a0Our approach has several advantages compared to standard\u00a0FISH: it enables one to target and label an individual\u00a0sequence element equivalent to the size of a PCR primer,\u00a0the small size of the probes reduces any effects that probe\u00a0incorporation has on the genome domain spatial arrangement,\u00a0and denaturation of genomic DNA is omitted. The\u00a0topological link between the target DNA and the circular\u00a0probe ensures that the precisely positioned label has to\u00a0occupy a fixed site along the double helix.
